Thirty four freshwater trout farms, in The Eastern Black Sea Region of Turkey were surveyed seasonally for bacterial pathogens and diseases between 2007 and 2008. And also a multiplex polymerase chain reaction (PCR) method was designed for five pathogens. A total of 558 fish were examined and 40 bacterial isolates were identified phenotypically. Yersiniosis, furunculosis, vibriosis, motile Aeromonas septicemia, bacterial cold water disease, and Pseudomonad infection were recorded. Infections caused by Yersinia ruckeri, Aeromonas hydrophila, A. salmonicida salmonicida, occurred most frequently, but only one and two outbreak of Pseudomonas putida, Flavobacterium psychrophilum and P. luteola infection were recorded, respectively. Most of the bacteria were isolated in spring and summer. Numbers of isolated bacteria were reduced in fall and winter. The disc susceptibility pattern was fairly consistent regardless of geographic areas or year of isolation. Fifty percent or more bacteria were resistant to ampicillin, cephalothin, erythromycin, neomycin, sulfamethoxazole, and tetracycline. The most effective antibiotics were oxolinic acid and florefenicol.A multiplex polymerase chain reaction (PCR) method was designed for the simultaneous detection of the five major fish pathogens, Aeromonas hydrophila, Aeromonas salmonicida subsp. salmonicida, Flavobacterium columnare, Renibacterium salmoninarum, and Yersinia ruckeri. The multiplex PCR assay was useful for the detection of the bacteria in naturally infected fish. This assay is a sensitive and specific and reproducible diagnostic tool for the simultaneous detection of five pathogenic bacteria that cause disease in fish. Therefore, it could be a useful alternative to the conventional culture based method.Key Words: Trout, bacteria, seasonality, disease, PCR
